Cloning of ACC Oxidase Gene cDNA from Tomato and Construction of Three Plant Expression Vectors

Abstract: Ethylene, a phytohormone, plays an extensive role in plant growth and development. The enzyme ACC oxidase, catalysing the last step in the biosynthesis of the plant hormone ethylene, is the key enzyme in the course. The main research of ethylene is to control the respiration amount through the method of antisense-RNA. In this research, a cDNA fragment of 1 018 bp was cloned using RT-PCR. Then, plant expression vectors with sense and antisense orientations and RNA interfering was constructed and transferred into Agrobacterium tumefaciens LBA4404 by electoral transformation methods.
