Abstract: To investigate the feasibility of diagnosis for nitrogen nutritional status of maize in molecular level, the elite hybrid variety Zhengdan958(Zea mays)was used to study the transcriptional difference of maize seedlings under different nitrogen levels by cDNA-SRAP markers.Several differentially expressed fragments were cloned and sequenced, and identified using Real-time PCR.The PCR amplification efficiency were various with different primer pairs. The nitrogen nutrition levels affected gene expression in maize. The results of cDNA-SRAP were basically consistent with those of Real-time PCR.Some fragments(such as N1,N9,N20,N30)could be taken as the candidate targets of molecular diagnosis of nitrogen nutrition in maize.cDNA-SRAP markers is an efficient way to develop molecular diagnosis of plant nutrition.
