Effects of Ultrasonication Treatment on Genetic Transformation by Pollen-Mediated in Apple

Abstract: The article discussed the effects of additional ultrasonication treatment on the germination of apple(Malus pumila Mill) pollen during pollen-mediated genetic transformation of apple cultivars Danxia and Gala. In order to find out suitable buffer solution and ultrasonication parameters, different pH values, cane sugar concentrations of buffer and various powers, treatment times, treatment number of ultrasonication were set up and examined through experiment. The results showed that the suitable buffer pH value fit for apple pollen treatment was 5.4 and the suitable concentration of cane sugar 10%. It was also indicated that such parameters as the power, time and treatment number of ultrasonication had significant effect on the broken rate and the germinated rate of apple pollen.
