Effects of Different Factors on Quality of Genomic DNA Extraction in Peanut

Abstract: To explore the factors on CTAB method of plant DNA extraction that affect the quantity and quality, seven methods phenol-chloroform, 10 000 r/min, chloroform, no cutting on peptips, one step with chloroform, one step with phenol, no shaking for 30 min have been used to extract genomie DNA from peanut leaves of Arachis hypogaea. The DNA samples obtained by above methods were examed by appearance, agarose gel electrophoresis and ultraviolet spectrophotometry. Based on the comparative analysis of yield and quality of the DNA samples by different methods, consideration all index of yield, protein content and the integrity of DNA molecules, the phenol-chloroform method was found to be the most suitable for extraction of genomic DNA from leaves of A rachis hypogaea.
