The Influence of the DifferentR ice Tissues TotalDNA in ChloroplastMarkersAnalysis

Abstract: "Chloroplast DNA marker is a common method of rice molecular analysis.However extract rice chloroplast DNA is a complex and time-consuming process.In order to simplify the steps of analyzing,this study use the CTAB and SDS methods to extract the total DNA(cultivation rice indica 93-11 and japonica SN265) from three different tissues of rice,including leafs roots and seeds,then amplify the total DNA templates with three pairs of chloroplast gene interval area markers(psbA-trnH,petG-trnP and infA-rpl36) and two pairs of chloroplast Indica-japonica classification markers(ORF100 and ORF29-TrnCGCA).The amplification results of roots and seeds are consistent with the leaf results and are also in expectation.And then use sixty weedy rice seed total DNA templates with chloroplast marks of Indica-japonica classification in PCR reaction.The results show these groups of chloroplast markers can amplify the target fragment from weedy rice seed total DNA and do Indica-japonica classification effectively.This conclusion can simplify the analysis process with rice chloroplast DNA markers and accelerate the analysis speed. "
