Transformation of Wheat with DNAj Gene from Foxtail Millet via Pollen-tube Pathway

Abstract: In order to obtain drought-tolerant transgenic wheat lines,DNAj gene from foxtail millet,which was demonstrated to be related with drought and heat tolerant,was tried to introduce into 4 wheat cultivars via the pollen-tube pathway.The effects on seed setting under different pollination methods,different treatments with stigmas,DNA concentration and DNA diluents were studied and only the method of pollination showed distinct influence on the seed setting rate.Genomic PCR of T0 generation plants revealed that the DNAj gene fromfoxtail millet was integrated into one wheat plant by repeated assay.This indicated it was feasible to introduce DNAj gene fromfoxtail millet into wheat via the pollen-tube pathway even the transgenic efficiency was only 1. 42 ‰.Our work has established the foundation for creating drought-tol-erant wheat materials with foxtail millet genes.
