Molecular Cloning of Granule Bound Starch Synthase II Partial Sequence and Its Construction of Antisense and RNAi Expression Vectors

Abstract: Partial cDNA sequences(1 069 bp)of granule bound starch synthase II(GBSSII)gene(GenBank No.EF221761)was amplified from grains of common wheat cultuvar Yujiao 2 by RT2PCR in the paper. The result demonstrat2ed that the cloned GBSSII gene sequence had 95 % - 97 % identity with the reported GBSSII genes in GenBank. In addi2tion, its antisense expression(pWM101G BSSIIA)vector was constructed with pMW101 vector, and RNAi vector(pFGC5941G BSSIIsa)was also constructed with pFGC5941. These constructed vectors would provide a good backgroundfor investigting the function of GBSSII on biosynthesis of starch in wheat plants.
