    The full length cry8Ea gene was amplified by PCR using a pair of special primers,JJX5 and JJX3,de-signed according to cry8-type genes sequences,and inserted into E. coli expression vector pET-21b to obtain the recom 2 binant plasmid.Nucleic acid sequence analysis showed that this gene was 3 495 base pairs encoding 1 164 amino acids, which were homologof 99.31 % compared with Cry8Ea1,the molecular weight of the protein was 131.8 kDa with isoelec-tric point pH4.71. This gene sequence had been registered in GenBank(accession number was EU047597),and named cry8Ea-as a novel gene by Bacillus thuringiensis Delta Endotoxin Nomenclature Committee. The result of SDS-PAGE in- dicated that cry8Ea-gene could be expressed as a 130 kDa protein in E. coli BL21 (DE3) strain induced by IPTG.Bioassay of the expression product from the cry8Ea-gene showed evident toxic to the larvae of Popillia flavosella-ta, Holotrichia parallela and Plagiodera versicolora,with the corrected mortality of 46.67 %,55 1 56 % and 33.33 %,re-spectively.

    By using the sequence of stripe virus published on line and the software of siRNA Target Finder, we ob - tained 168 siRNA segments.After BLAST Scan of rice genome, 6 segments were selected and 2 were chemically synthe - sized and used to construct of expression vector ( pASV1301 -1, pASV1301 -2) intergratedwith pCAMBIA1301 plasmid.The recombined plasmid was successfully transformed into agrobacterium of EHA105,which lay a foundation for the transfor - mation of rice and the prevention of stripe virus disease of rice by RNA interference.

    One pair of primers was designed based on the pig IL- 2 gene sequences published in GenBank and used to amplify the interleukin-2 cDNA of Huainan Pig by RT-PCR from total RNA extracted from spleen lymphocytes,which were cultivated and stimulated with 10 μ g/mL ConA in vitro for 27 h. Then the amplified cDNA was cloned into pGEM- T Easy vector and sequenced. The result showed that the full length of Huainan pig IL- 2 was 516 bp with an ORF of 465 bp,encoding a peptide of 154 amino acids which molecular weight is 17. 4 kDa, pI 5. 27, hydrophobic amino acid 38. 3%, hydrophilic amino acid 42. 0%,basic amino acid 11%and acidic amino acid 23%. The sequence was the same as the corresponding genes published in Genebank. The base similarity of pig IL- 2 with those of cat,cattle,chicken,dog, duck,goat,horse,human and house mouse was 83. 7%,82. 6%,28. 2%,80. 6%,29. 6%,83. 4%,81. 3%,82. 0%and 61. 5%, respectively.

    One pair of primers for amplifying the F gene of NDV on GenBank were designed,withwhich the predict - ed 1. 7 kb fragment had been amplified via RT -PCR from NDV strainHebei. The 1. 7 kb fragment was inserted into pMD - 19T vector. DNA sequence analysis showed that the cloned fragment consisted of 1662 bp and coding for 553 amino acid. Compaired with 13 strainsof NDV on GenBank, neucleitide and amino acid sequence homologieswere found to be 97. 2% - 99. 2% and 96. 9% - 98. 7% respectively, but the amino acid sequence homologies to other NDV strains such as La - Sota,F48E9 and B1 were only 88. 6% - 91. 5%. The deduced amino acid sequence of the cleavage site of F protein of the NDV isolates were 112R -R -Q -K -R -F -I -G119. the NDV isolates belonged to the genetype.

    The disruptive effects of Fluorescent Brightener 28( FB28) and Heliothis armigera granulovirus enhancing ( En -Ha) on the peritrophic membrane( PM) of larvae and enhancing effectsonMsNPV in Mythima separata were studied through SDS -PAGE, structural observation and bioassay.The results indicated that proteins could be dissociated from the PM in vitro by FB28.After the larvae were reared with diets containing 1% FB28 for 4 h, bluish fluorescence appeared on the disrupted PM.SDS -PAGE of PM treatedwith FB28 in vivo showed that some proteins with high molecular weight disappeared.After the PM was treated with En -Ha in vitro, some proteins with high molecular weight disappeared and some with lower molecular weight appeared, which indicated En -Ha could degrade the high -molecular -weight proteins from the PM.The PM became fragile after the larvaewere rearedwith diets containing 9μg/mL En -Ha for 7h, and SDS -PAGE showed that some proteinswith high molecular weight disappeared similar to the profile treated by FB28?However, the ex - periments testified that the disruptive effect could be restored after rearing the larvae on the diet without FB28 and En - Ha.The bioassay showed that FB28 and En -Ha could enhance the vitality of MsNPV and significantly decreased LC 50 by degrading the integrity of PM.

    DNA methylation is known to play an important role in the regulation of gene expression in eukaryotes. The wheat near-isogenic line TcLr19 and Thatcher is race-specific resistance and compatible in the interaction with Puccinia triticina respectively. Using methylation -sensitive amplified polymorphism( MSAP)analysis,we compared the patterns of cy- tosine methylation in TcLr19 and Thatcher that had been inoculated with Puccinia triticina,subjected to left untreated. In all,3 554 DNA fragments,each representing a recognition site cleaved by either or both of two isoschizomers,were amplified using 60 pairs of selective primers. A total of 998 sites were found to be one of the methylated. The results suggest that no differences in cytosine methylation exist between seedlings inoculated and those unchallenged.

    In order to offer the theoretical data for prevent and control PRRS that unparalleled outbreak in Hebei. According to the reported complete nucleotide sequence of PRRSV CH-1a and JXA1 inGeneBank,3 pairsof primers were designed to amplify ORF5-7 gene of HB -4( hs)strain with Primer 5. 0 software. The ORF5-7 gene nucleotide se- quenceswere obtained by RT-PCR and sequenced after being cloned directly into the pGM-T vector. The obtained sequences were analyzed and compared with the published complete nucleotide sequences of other strains in GeneBank by DNAstar software. The results showed that the PRRSV HB-4( hs)strainwas clustered within North American genotopy. The ORF5-7 gene of HB- 4( hs)strain showed highest homology of 98. 8%- 99. 5%to the strains were isolated in recent years,showed 91. 9%- 92. 6%identity with the strainswere isolated in last years. The phylogenetic trees revealed the strains were isolat- ed in recent years had close relationshipwith HB -1( sh)strain.

    The cross of watermelon PI482246,which is resistant to powdery mildew disease, and Zhongxin 1,which is susceptible to powdery mildew disease, were used to construct F2, BC1.1 and BC1.2progeny populations?The inheritance of resistancewas analyzed with Podosphaera xanthii race-France purified and identified from a collection of powdery mildew using spore spraying method in greenhouse. The results of cross tests indicated that the resistance carried by PI482246 to powdery mildew race-France was controlled by a single recessive gene and the resistance was incomplete recessive.

    In the study, the allelopathy of the ethanol extract of the root of parsley on Fusarium oxysporiumf. sp. cuc- umeris was studied by growth rate method.The result showed that the ethanol extract of the root of parsley inhibited the growth of Fusarium oxysporiumf. sp. cucumeris significantly within the range of experimental concentration except the con - centration of 1% in 120 and 132 h, and its highest inhibitory rate was 42.31%,or lowest rate was 12.22%;the ethanol extract of the rhizosporic soil of parsley inhibited the growth of Fusarium oxysporiumf. sp. cucumeris significantly at higher concentrations( from RS100% to RS60% ), and the largest inhibitory rate was 94.3%.The higher concentration, the bet - ter allelopathic effect, whichwas more than that of corresponding extract of the nonrhizosphere, adding up to 49.02%; the lower concentration( RS40% and RS20%) of the ethanol extract of the rhizosperic soil of parsley promoted the mycelial growth lightly, but whichwas less than that of corresponding extract of the nonrhizosphere obviously.There was a tendency that inhibitory rate of ethanol extract of root agaist Fusarium oxysporiumf.sp. cucumeris dropped down with decreasing concentration and prolong time.

    The behind fragment of PCV2 ORF2 gene was amplified from the extracted DNAof PCV2 in inoculated in PK-15 cells;The PCR product was connected with pMD19-T vector,which was confirmed by sequence analysis and re-strict enzyme identification,and then was cloned into pET- 32aVector,Plasmid containing the right insertion was digested with enzymes to confirm its identity and retransformed the recombinant plasmid into E. coli BL21 strain. The bacteria were induced by IPTGand the lysates were analysed directly by SDS-PAGE.Results were asfollows. Afusion protain ap-proximately 40 kDa in molecular weight was observed on the SDS-PAGE. Western-blotting confirmed that had antigenic activity of PCV2.

    As an initial step to study the structure of PM from Holotrichia oblita, the basic structure of the PM was analyzed by microscopy and the PM morphological aspects were observed by biochemical technology. The results showed that major components of the PM were proteins accounting for 36.25%, and carbohydrates accounting for 8.91% by weight besides chintin.PM surface appeared to be smooth, close and highly elastic. Apparently 28 polypetides were sepa - rated by SDS -PAGE analysis with molecular weight from 17.6 to 320. 3 kDa. Heliothis armigera granulosis virus en - hancin could not degraded any proteins by assay in vitro.

    In order to study effect of the gene SeNHX1 on yeast tolerance to salt, vector pYES2 - SeNHX1 -GFP、 pYES2 - SeNHX1 and pYES2 - GFP was constructed and transformed into yeast W303.Transformed W303 was cultured in CM -ura medium with D -gal which induced the vector expressing in Yeast1Bright green under fluorescence microscopewas observed in W303( pYES2 -SeNHX1 -GFP) and W303( pYES2 - GFP), but not in W303( pYES2 -SeNHX1).W303( pYES2 - SeNHX1 -GFP) and W303( pYES2 - SeNHX1) grew more slowly than W303( pYES2 - GFP) in 0- 48 h, it grew fast after 48 h1It indicated that the vector expressed correctly in yeast and gene SeNHX1 could enhance the tolerance to salt.

    In order to research FUT I genecs effect to the growth performance of piglets, the polymorphisms at M307 of FUT I gene inLaiwu Black pigs, Duroc and Yorkshire were investigated and their associationswith growth performance were analyzed.The result showed that there were polymorphisms at M307 of FUT I gene in Laiwu Black pigs, Duroc and Yorkshire, and the populations were all not in a state of Hardy -Weinberg equilibrium( p< 0101) 1G allelic frequency was the most in Laiwu Black pigs, while in Duroc and Yorkshire, the A allelic frequency were the most. The three genotypes had significant effect to Duroc ( p< 0105) and Yorkshire ( p< 0101) and no significant effect to Laiwu Black pigs on weight at day 70th,which is closely related with the diarrhea and edema of weaned piglets1It was inferred that because of the difference of breeds, FUT I can be an effective candidate gene for ETEC F18 to Duroc and Yorkshire, but not an ef - fective candidate gene to Laiwu Black pigs1Further study should be done to identify and characterize putative QTL and functional gene responsible for the resistance to ETEC F18 in Laiwu Black pigs.

    For G lutathion-Peroxidase 5 ( GPX5 ),Fucosyltransferasel ( FUT1 ) and Nuclear receptor co-activator 1 ( NCOA 1) gene,the target segment of those genes were amplified through PCR,and then these amplified segments in five strains of pig were genotyped by PCR-RFLP 1 The strains involved were Landrace Y orkshire,Duroc,Min pig and Crossbred pig.After genotyping by PCR-RFLP with Hinf lin GPX5 gene obtained 3 genotypes;With Hha lin FUT1 gene ob-tained 3 genotypes;With Rsa lin NCOA1 gene obtained 3 genotypes 1 Then the frequency of gene and genotype of these genes were calculated,and analysised for genetic effect.

    In this study, Triticum aestivum L 1 9-Cis-Epoxycarotenoid Dioxygenase gene ( TaNCED1 ) was amplified using specific primers containing restriction enzyme site of Kpn I and Xba I.PCR products and the binary expression vector pCAMBIA2300 ~ 35Swere digested by the corresponding restricted enzymes respectively,and linked directionally.The re-sulting construction with TaNCED1 was named pTaNCED1.The pTaNCED1 was further introduced into Agrobacterium tumefaciens strain G V3101 for the following-up research such as gene functional identification and gene transformation for wheat stress tolerance improvement.

This paper uses the method of the transformation expression of gus gene and PCRmeasure, de - tected 68 different materials of mature embryos in transformation expression of gus and 8 different materials of plants which were transformed from immature embryos, the results showed that foreign genes can be detect in Yangmai10, Shilu - an02 -1, Shijiangzhuang5, Zhoumai18, Shangdong01 -35, Taishan008, Y9 -63and Handan3475, so they are sensitive geno - type materials and can be used for the receptor of the Agrobacterium infection; Gus genes were expressed in Yangmai10 and Shiluan02 -1, the best effects of infection were resuming training in 14 The embryos members of Gus gene expres - sion were increased, and the volume of locus caeruleus were augmented?So Yangmai10 and Shiluan02 -1 can be used for the materials of the grading -up system.

    The expression of gene fragments in the corn leaves were studied both in incompatible combination(B37 Ht1 inoculated with race 0 of Setosphaeria turcica ) and in compatible combination(B37 Ht1 inoculated with race 1 of Setosphaeria turcica ) by mRNA differential display PCR.The results showed that 17 gene fragments were expressed ex-clusively,26 gene fragments expressed increasingly,and 20 gene fragments expressed decreasingly in incompatible combi-nation compared with either in compatible combination or in control. Homology was not found between gene fragments ex-pressed exclusively in incompatible combination and known R genes.

    Gene mutation ratio could be improvedby irradiation.To study the rote of irradiation techniques in the rela -tionship between irradiation dosage and generation after irradiation is important?3 cultivars of soybean were irradiated by 6 irradiation dosages of 60 Co -rays, 50, 100, 150, 200, 250, 300 Gy and 0 Gy( ck), respectively, and the ratio of irradiation dosage was 1.2Gy/ m in this paper.Based on the living plant ratio inM 1 and the mutation effect inM 2, the best irradiation dosage and the irradiation dosage in M2were evaluated in the test.The results showed that the mean value of the agronomic characters which were control growth was in negative correlation with the irradiation dosage in the M1 generation by 60 Co -rays.The equation follows as y= 0.0001x2 - 0?381x+ 102.9( R 2 = 0962 7**) ;The best irradiation dosage was different in cultivars andtheir mean value was 144.3 Gy; and the plant height, mainnode number, pod length, pod and grain number per plant and hundred -grain -weight were affected by different irradiation dosage in M2, and the more irradiation dosage, the greater effect in a certain extent; gene mutation enabled the plants to exceed their parents in M-generation.

    This paper identified the chromosomal ploidy of rice according to the pollen,plant morphology,seed mor-phology and chromosome number.The results showed that the size of plant cells and organs tends to be enlarged with the increase of chromosomal ploidy .It was concluded that the size of pollen in rice has a positive correlation with the chromo-some number and ploidy,while the plant morphology,grain size,availability of awn,grain density,setting percentage and other traits also have some correlation with the chromosomal ploidy.They can be a reference to distinguish the chromoso-mal ploidy of rice.

    Genetic relationship among four types cucumber germplasms was analyzed by AFLP markers?To make up the systematic diagram of genetic relationship by NTSYS software and clustered by UPGMA method.24 pairs of AFLP primers had been selected from about 50 pairs of AFLP primers.Result indicated 1 367 bands were amplified with 24 pairs of primers and 297 polymorphic loci were found which accounted for 21.7%.Genetic similarity coefficient were changed from 0.841 3 to 0.994 9.By cluster analysis, twenty -eight accessions were clustered into seven groups, different germplasms diversity of HUABEI cucumber is lower than the other three types cucumber germplasms.It showed that AFLP were an effective, repeatable and dependable marker for polymorphism analysis in cucumber germplasms.

    Biological characters of 79 asymmetric somatic hybrids obtained through protoplast fusion between Brassi- ca oleracea L1var1botrytis( 2n= 18, CC) and Brassica nigra ( 2n= 16, BB) were studied1The results indicated that the morphology of hybridswere heterogeneous1Most of the population was intermediate to that of the parents, compared with minority of them were similar to parents1Nuclear DNA content analysis revealed that hybrid plants had different nuclear DNA content: higher, lower than the parental sum or equal to it1Contrast to the lower and equal hybrids, bigger amount of hybrids with higher DNA content had abnormal leaf, bad growth vigour and poor fertility1It showed that growth vigour and fertility had relations with the nuclear DNA content1By chromosome counting, the detected hybrids were mixoploid1Among 79 hybrids, 13 aimed hybrids were picked out,which were vigorous and fertile. Broaden the genetic variability of cabbage though asymmetric somatic hybridization was indicated feasible.

    In order to enrich the germplasm resources of B. campestris, crosses between Brassica. campestris L. pekinensis and B. nigra were made and thirteen hybrids were produced. Three methods including morphology,cytology and SRAP (sequence-related amplified polymorphism) molecular makers were used to verify the hybrids 1 The pollen stain-ability and fertility of the hybrid plants were also analysed.Morphologically the hybrids showed an intermediate phenotype while the dendrogramon SRAP data indicated that the hybrids were homophylic to the female parent. The pollen stainabil-ity of the hybrids was low,among which only one(H4)could produce seeds after backcrossed to the B campestris. Some of the cells in the hybrid H4 had 18 chromosomes while 48 percent of cells contained 26 chromosomes;The other twelve sterility hybrids had 18 or less than 18 chromosomes. Results indicated that the fertility of the hybrids was related to the chromosome number and the increased number of chromosomes could enhance fertility.

    In this study,by PCR amplification with the genomic DNA of three mapping parents,940 SSRs primers were tested for analysis their versatility and polymorphism in melon,cucumber and watermelon. The results indicated that these SSR primers had some versatility in the above crops. The minimum SSR transferability was higher than 24 %,show-ing some similarity in the genome of these crops.But these SSR primers also had strongly genus specialty,their polymor-phism level was very low,the ratio of common polymorphism SSR primers among these crops was lower than 1 %.

    In this experiment,during the process of goat oocyte IVM,by adding different concentration cumulus cell and its surrounding granular cell,we studied the inhibitive effects and its mechanism of follicular cells on meiosis of oocytes. The IVM rate of oocyte was not being affected considerably after adding 10 5 and 106cell/ mL cumulus cells or surrounding granular cells,contrast to control group (p > 0.05),but were significantly higher than that of 5 × 106cell/ mL(41.2 %),107 cell/ mL(24.6 %)cumulus cells and 5 × 106 cell/ mL(38.5 %),107 cell/ mL(23.8 %) surrounding granu-lar cells(p < 0.05). In adding 5 × 106,107cell/ mL cumulus cells group and surrounding granular cells group,the cumu-lus could not expand.Nuclear Status Examination,the meiosis of oocytes was maintained in G V stage. Rp-cyclic adenosine monophosphothioate,a membrane permeable antagonist to cAMP,induced the oocyte IVM rate of 107cell/ mL cumulus cells group (71.6 %)and surrounding granular cells group (71.3 %)which there was no significant difference among the control group (p > 0.05). during the process of oocyte IVM,adding some concentration cumulus cells and surrounding granular cells would maintain meiotic arrest by increasing the cAMP level of oocyte,make oocyte stay in the G V stage.

    The experiment was conducted during 2006 - 2007 in field conditions on irrigation and Nitrogen Coupling effects on nitrate reductase activity,plant nitrogen absorption and yield of two different types of winter wheat Shixin733 and Shimai15.The result showed that the nitrate reductase activity of Shixin733 increased with the nitrogen amount,but the nitrate reductase activity of Shimai15 had the highest point among 180 - 240 kg nitrogen amount per hectare,de- creased when too much or too less nitrogen. The nitrate reductase activity decreased when the nitrogen was too much or too less on the high soil containing water,increased with the increasing nitrogen on the low soil water content.Proper nitroge- nous fertilizer could promote nitrogen absorption of the plant,but the anthesis irrigation improved the nitrate reductase ac- tivity of the flag leaves and grain yield,but remarkably reduced the whole nitrogen content of the grain and had less effec- tion on the whole nitrogen absortption of the plant On the experimental conditions,Shixin 733 had the highest yield when it was irrigated two times and feilizered 180 kg per hectare,Shimai 15 had the highest yield when it was irrigated two times and feilizered 60 kg per hectare.

    As a second messenger, Ca2+ plays an important role during the plant resistance to biotic or non -biotic stress.And as a carrier of Ca2+, Ca2+ -ATPase intervenes in many reactive processes of outer stimulation, so as to keep steady -state [ Ca2+] cyt level. In the report, Ca 2+ -ATPase activities of microsomal membrane system, including cytoplast membrane, mitochondrion membrane and vacuole membrane,were measured under osmotic stress with PEG6000.The re - sults showed that the Ca2+ -ATPase activities of normal plant changed with the longer light from 7: 00 AM to 7: 00 PM?In generally, the total Ca2+ -ATPase activities of three membranes were close and changed while plant subjected to osmotic stress, and it's higher in leaves than that in roots both treatment and control. In leaves, the Ca2+ -ATPase activities were decreased while treated with PEG, ABA and EGTA could reverse the effect, but it changed little in root while subjected to PEG,and were all increased by outer reagents.In detail, the Ca2+ -ATPase activities of three membranes were all in - creased by PEG in leaves, and so as to both mitochondrion membrane and vocuole membrane in root. But the Ca2+ - ATPase of cytoplast was inhibited obviously by PEG. Four outer reagents had different effects on Ca2+ -ATPase activity.In leaves, the decrease of Ca2+ -ATPase by PEG could be reversed by both ABA and EGTA, and the activities were higher than that of control at 6 h after treatment by PEG by two reagents?But the effects were different in roots.Vp could re - versed the decrease by PEG in cytoplast membrane, and increased Ca2+ -ATPase activities of mitochondrion membrane, and the vacuole membrane Ca2+ -ATPase activity were increased by ABA obviously.Different from those,TFP could pro - mote the Ca2+ -ATPase activity to raise in prophase of osmotic stress, but inhibited it after 6 h while treatment with PEG. Those indicated that three membrane both leaves and roots were all participate in the regulating process of keeping low [Ca2+ ] cyt under osmotic stress.External reagents influenced Ca2+ -ATPase activities both leaves and roots, but the re -sponse differences existed between leaves and roots.

    The effects of irrigation management on chlorophyll(Chl)content of flag leaf were analyzed in a field study for six high-yielding winter wheat cultivars. The results showed that soil water deficit (0 - 100 cm,RSWC 40 % - 60 %) had little effect on Chl content of flag leaf during 0-14 d after anthesis,but accelerated Chl degradation at late grain filling stage. RSWC higher than 70 %of the 4 2 time- irrigation decreased Chl content and Chla/ b,and slowed the degradation rate of Chl,which treatment resulted in earlier Chl content peak compared with the other treatments.RSWC range of 60 % - 70 % was favorable for higher Chl content. Cultivars of SX733,SX703,SM15 and SM12 were sensitive to irrigation in terns of Chl content,but JM5579 and SJZ8 responded reversely.No significant correlation was found between mean post- anthesis Chl content of flag leaves and grain yield among different irrigation treatments.Stronger positive correlation rela- tions were,however,revealed between 28 d Chl content of flag leaves and grain yield.

    In this study Longdong( strong drough- t resistance) Gongnong No 1( medium drough- t resistance) and BL -02 - 329( weak drough- t resistance) were chosen as experimental material to study the change of water metabolism and two os - motic adjustment substances in the leaves of alfalfa under drought stress.The results showed: the water potential and water relative content in the leaves of alfalfa decreased and water saturated deficit in the leaves of alfalfa increased under drought stress.There were higher decrease extent of water potential and lower decrease extent of water relative content in the leaves of strong drough- t resistance alfalfa under drought stress. At the same time the proline content and soluble sugar content increased in the leaves of three alfalfa, the two substances both have adjustive function to three alfalfa under drought stress. Proline is the main adjustment substance and the content and the increase extent can reveal drough- t resis - tant of alfalfa cultivars.

    This paper using the summer maize hybrids Zhengdan958 as the experimental material,the effects of dif 2 ferent concentrations of KCl on the aging seeds germination of maize and some physiological characteristics including the activities of antioxidase and dehydrogenase,the contents of malondialdehyde (MDA),soluble sugar and soluble protein were analyzed.The results showed that 0. 1 - 10.0 mmol/L KCl treatments could increase the germination potential,the vigor index and dry weight of seedlings of maize .It was also observed that the activities of antioxidase and dehydrogenase and the content of soluble sugar and soluble protein increased,but the content of MDA decreased.The results indicated that KCltreatments enhanced antioxidation level,mitigated lipid peroxidation,maintenanced the integrity of plasma mem-brane and improved substance metabolism rate of germinating seeds,and this might be the physiological mechanismof the aging maize seeds germination accelerated by potassium.

    Effects of NaCl stress in different concentration on growth and photosynthetic characteristics in leaves of lettuce seedlings were studied.The results showed that the plant height and root length were inhibited obviously.The con- tent of chlorophyll decreased with the NaCl concentration and treated time.However,it increased under 50 mmol/L NaCl. The content of chlorophyll a dropped more rapidly than that of chlorophyll b. The net photosynthetic rate (Pn),stomatal conductance(Gs)and transpiration rate(Tr)changed similarly with increasing salt stress,and all decreased.Stomatal con- ductance and intercellular concentrations of CO2 (Ci) changed similarly under 50 mmol/L NaCl stress.The changes of stomatal conductance were contrary to intercellular concentrations of CO2 under 100 - 200 mmol/L NaCl. It was stomatal limitation that leading the photosynthesis declined in the treatments with lower concentrations.On the contrary,non-stom- atal limitation was the dominative causation in the treatments with higher concentrations.

    The leaf length,the leaf width and the shoot length were studied under five moisture treatmentswhich were SWC30,SWC45,SWC60,SWC75 and SWC90 treatment of red soil in Spring. The characteristics and content change of N,P and K element in leaves and roots of orange were also compared under those soil water treatments. The result was showed that it was better for leaf growth of orange when soil water treatment was about 75%. There was no difference for N in leaves and roots. But the contents of P and K in leaves were less than that in roots. The appropriate water shortage was advantageous to N cumulation for orange. The content of P in orange leaveshad a positively significant correlationwith soil relative water content( SWC). There was no difference for the content of K in leaves among different SWC treatments.

    The NaCl stress simulating technology at seedling stage in greenhouse was used for the studies on the ef- fects of salinity and days treatments on physiological characteristics,such as the relative water content(RWC),cell mem- brance permeability(CMP),content of free proline (Pro) and Malondaildehyde (MDA)of 13 germplasm and materials of E.intermedia (Host)Nevski collecting from 12 different countries.The results showed that the RWC was decreased,but the CMP,Pro and MDA were increased when the stress of salinity and days were intensified.According to the correlation and cluster analysis of 4 physiological indexes,13 germplasm and materials of E.intermedia were divided into 3 levels of salt tolerance,e.g. the strong salt tolerance of EI037,EI038,EI043,the weak EI020,EI030,EI041,EI042,EI045,and the medium EI017,EI031,EI044,EI046,EI049.Furthermore,0.9 %of NaCl concentration was considered as the salinity threshold of E.intermedia germplasm at seedling stage based on above 4 physiological indexes,the salinity tolerance rat- ings and livability.

    Gas chromatography was used to analyze the alkaloid composition and content of air-cured burley leaves from different areas and varieties; the proportion of converters and the conversion rate were calculated. The results indicat- ed that the nicotine conversion level in Sichuan burley tobacco was significantly higher than that in other producing ar- eas. The leading variety Xuanhan-5had serious problem of nicotine conversion,especially for the narrow-leaf type which is currently the most popular variety,with the proportion of converters of 96% and the average conversion rate of 34. 8%. The broad leaf type Xuanhan- 5 also had high conversion levels. Hybrid Dabai had relatively less problem of nicotine conversion, the proportion of converters in 3 locations being 18. 0%, 18. 0% and 52. 0%,respectively, and the average conversion rate being 3. 5%,3. 1%and 14. 3%,which can be used after screening and improvement. In the population of Eyan 1 and TN90,some converterswere also found at different levels. The newly developed hybrids Dabai 3 and Dasuo 24 had serious problems of nicotine conversion,with a converter proportion of 100%, thus the systemic selections and im- provementswere required. The variation of nicotine conversion was found in different locations for the same variety.

    he physiolygical mechamisam study of ultradrying and aging sunflower seeds during germinating showed that fresh weight,germination percent,germination index and vigor indexof ultradried sunflower seeds were significant dif- ferent from ultradried after being aged;the ultradrying storage helped to keep the vigor of sunflower seeds and especially all indexes of ultradried sunflower seeds(MC = 2. 08 %)were improved most. With the germinatingof seeds,the content of heat-stable proteins decreased;MDA content of ultra-dried sunflower seeds were lower than that of unultradried ones;anti- oxidation enzyme systemof the seeds(MC = 2. 08 %)retained best,which alleviated or prevented the process of the super- oxidation of the inner membrane lipids started by active oxygen,avoided the harm to membrane system,ensured that seeds germinated smoothly.

    To protect the germplasm resource of wild Allium mongolicum, prove up the ability for bearing the salt and the feasible condition of domesticating planting. This paper studied the effect of NaCl stress on vigor and activities of an - tioxidant enzymes in Allium mongolicum seed, which the treatments are different salt concentrations. The results showed that:With the increasing of salt concentrations from 20 to 500 mmol/ L, the germination rate, all of germination energy and germination index was decreased significantly except of 20mmol/ L salt treatment, the salt injury ratio of every treatments was increased 13.7%- 99.0% than the control.The SOD and POD activities were significantly higher than the control under 20 and 50mmol/ L salt treatments, but lower when the salt concentrations is above of 100 mmol/ L.The CAT activ- i ty was increased gradually with the increasing of salt concentration. Under NaCl treatments with 20 and 50 mmol/ L, the content of MDA of Allium mongolicum R.Seed is a little lower than that of the control, but increasedwith the increased of the salt concentrations above of 100mmol/L.And the MDA contents of every treatments were all higher than the control. According to above,The Allium mongolicum R. seeds adapt to bourgeon under the salt treatments lower than 100mmol/L.

    The effect of different storage temperature( 17, 12, 7 ) on the main quality index of cucumber seedlings during storage was studied.The result showed that below 12 and 17, the two physiological indexes( activity of root vigor and chlorophyll content) dropped less, the MDA and Pro rose less, the seedlings had the greatest dry weight, the growth above and underground kept balance, and the growth speed of the central leaves was faster after transplanting. But the seedlings stored in 12 grew better than in 17 after 6 d of storage.

    For make the reasonable measures of florescence control,this article reveals the physiological characteris- tics of Syringa oblata flower bud differentiation. Changes of soluble protein,soluble sugar,starch and nucleic acid con- tents in floral bud,leaf bud and leaves of Syringa oblata during its floral bud differentiation were studied. The results are as follows,in the physiological differentiation stage,the contents of soluble sugar,soluble protein,starch and nucleic acid accumulated highest in floral bud,but the soluble sugar accumulation in floral bud is much more than that of the leaf bud 1 It results such index are helpful to the formation of floral buds.After the contents of soluble sugar,soluble protein, starch and nucleic acid of floral bud reduced in the floral bud shape differentiation,they are rising continually,and they are higher than leaf bud during the floral bud differentiation.

    Using 3414 quadratic regression optimum design, the effects of N, P, K mixed fertilizing on fresh forage yield and nutriention of Argangjing alfalfa( Medicago sativa cv.Argangjing)was studied in gray meadow soil of West Liao Rever Plain of Inner Mengoulia Autonomous Region in 2003.The result showed that the optimum combination of N, P, K wasN: 3.37 kg/ hm2, P2O5 : 55?61 kg/ hm 2, K2 O: 256.63 kg/ hm2and the fresh forage yield of alfalfa was 77 944 kg/ hm2. In the 14 treatments, the contents of crude nitrogen of treatment 8 and treatment 10 were higher, and the contents of crude fiber in the treatment 7 and treatment 8 were low down 25% at early blooming stage and the contents of crude protein and crude fiber reached the 1st level of the state's standards.The content of crude ash was lower in the treatment 8 at early blooming stage.This study gived a rational fertilizing program for planting Argangjing alfalfa in theTongliao area, achieving high quality forage on the basis of increase yield.

    Eight years 'location experiment was conducted in cinnamon soil in Hebei province to study the distribu- tions of organic carbon and nitrogen in soil and its micro-aggregates,and to study the changes of the soil organic carbon and nitrogen fertility under different fertilization regimes from the view points of the storage of organic carbon and total N in various micro-aggregates. The results showed that the composition of soil micro-aggregates is different under different fertilization regimes. No fertilization and application the constant NPK fertilizers mainly decrease the storage of organic carbon and nitrogen in 10- 50 μ m, and resulted in soil fertility decrease. Application the increment NPK fertilizers can maintain the soil fertility,but the input ishigh. Application organic manure combined with constant NPK fertilizers mainly decreases the storage of organic carbon and nitrogen in bigger sizes,especially in 10-50 μ m micro-aggregates,and im- proves soil fertility. And it is the most effective way to improve soil fertility.

    Phosphorus and potassium morphology, content and accumulation characteristic of greenhouse were studied to distinguish the present situation of successive and excessive fertilization in greenhouse, using the means of comparison sampling in greenhouse, open vegetable field and grain -cotton field. The results showed that both phosphorus and potass- i um content had obviously accumulation in the 0- 25 and 25- 50 cm layers of greenhouse, and the accumulation of phos - phorus was higher than that of potassium, and the accumulation was higher and higher with protected cultivation age of greenhouse.Recent years the potassium accumulation in soil had the quickening tendency.The study on different morpho- l ogy and content of inorganic phosphorus showed that the proportion of Ca2 -P, Ca 8 -P in greenhouse was higher than that in open vegetable field and in grain -cotton field, and that in open vegetable field preceded that in grain - cotton field.The same tendency was found in slow available potassium content?Those showed that the accumulation phosphorus and potas -sium still had high validity.

    Soil enzyme activities were studied in soil planted with transgenic and notransgenic cotton under field con- ditions. The enzyme activities of urease,phosphatase and invertase were detected at seedling stage,squaring stage,flower and boll stage,boll-opening stage and senescence stage. The results showed that there were some different effects on soil enzyme activities due to different lines,growth stages and soil enzyme,but the same results were obtained at flower and boll stage and senescence stage. Two transgenic cottons planting significantly restrained activities of urease,while in- creased activities of phosphatase at the flower and boll stage significantly,and restrained activities of phosphatase and in- vertase at senescence stage significantly.

    Residue of NO 3 - -N in soil by fertilizer application under site -specific fertilization experiment about 19 years in Northwestern Shanxi Province was carried out.Results showned: NO3 --N wasn't accumulate in the soil with low organic fertilizer application, and appear linear type in soil profile distribution."S"typewas appeared of NO3 - -N accumu -lation in soil under other fertilizer treatments.Two NO3 - -N accumulation layer were shown in 20- 80 and 140- 200 cm,and formed NO3 - -N lower accumulation area in 80- 140 cm. Nitrogen fertilizer improved NO3 - -N accumulation on a large scale. However, combined phosphorus fertilization reduced NO3 --N accumulation significantly; accumulation of NO3 --N was about the same as application of nitrogen fertilizer combined with low amount fertilizer; accumulation of NO3 --N in soil rised with application of high amount of organic fertilizer.

    The effects of continuous cropping and rotation system on soil nutrient and soil microflora of tobacco field were studied,and the changes of nitrate nitrogen at different growth stages of flue-cured tobacco with different ratios of ni- trate nitrogen and ammonium nitrogen as base-fertilizer were analyzed 1 The result showed that,comparing to rotation pattern, the 3-year and 6-year continuous cropping tobacco led to the decrease of soil organic matter,the increase of soil available nitrogen,phosphorus and potassium,and the rise of pH.Compared to 3-year continuous cropping,the soil of 6-year continu- ous cropping increased more in pH,available phosphorus and potassium,had the similar level of available N,and decreased in organic matter by 4.3 %.The number of bacteria and actinomyces in soil presented a desending trend with the increase of continuous cropping years.The number of fungi was the lowest in 3-year continuous cropping. The number of effective mi- croorganisms such as bacteria and fungi reduced gradually along with the extension of continuous cropping years,while the number of harmful microorganisms such as pythium increased.The content of nitrate nitrogen in tilth layer increased with an increase of proportion of nitrate nitrogen in base fertilizer during early growth stage.The nitrate nitrogen mainly accumulated in 40 - 60 cm depth of soil as the development of tobacco.Based on the changes of nitrate nitrogen in soil during whole stage of growth,the optimum ratio of nitrate nitrogen in base fertilizer was 30 %- 50 %.

    By simulating acid rain and dus- t fall under the condition of green -house, spinach and lettuce were given different trial treatments in pot planting experiments.The results showed that the contents of Cu, Pb, Cd, Cr in spinach and in lettuce increased with the pH value decreasing through single acid effect, and the contents had a significant nega - tive relationship with the pH value.The contents of Cu, Pb, Cd, Cr in spinach and in lettuce were higher than the control test through single dus- t fall effect. And two vegetables showed different trends with the same heavy mental through the compound effect.For example, the contents of Cu in spinach increased in earlier stage and decreased in later as the pH value decreasing, but the contents of Cu,Pb, Cd, Cr increased in lettuce?For different heavy mental, the same vegetable had different pH acute points through the compound effect, such as spinach, the acute point of both Pb and Cd was pH= 4,while the acute point of both Cu and Cr was pH= 5. Compared to the food hygiene standard of PRC, both single and compound effect made the contents of Cd far beyondthe level, even the most multiple of exceeding standard was 2.5, then followed by Cr and Pb the last Spinach was acuter than lettuce for the sensitivity of acid rain.

    To detect the effects of different cytoplasmic background on bread -making quality properties of hybrid wheat,32 crosses from 4 strong gluten and 4 mid/weak glutenwheat cultivars based on NC II design,and 14 crosses from cultivarswith T-,K-,AL- and A-type cytoplasm were made, respectively. F-seeds of each cross were harvested and used to test the hardness, GMP content, Zeleny-sedimentation value, Farinograph parameters and Loaf volume. The results showed that the strong-gluten cytoplasm( Yumai34,Zhengmai9023 and Shanyou225) had positive effects on all bread-mak- ing quality properties,except water absorption and Valorimeter value,so the strong gluten wheat cultivar should be used as female parent in the program of bread wheat breeding. The T-and K-type CMS cytoplasms had general positive effect on Loaf volume,compared to A-type and AL-type CMS cytoplasm. The effect of CMS cytoplasm on loaf volume for crosses T- XN772× AL-R1 and K-XN772× T6 -3 reached to 60 and 98 mL, respectively. The T-,K- and AL-type CMS cytoplasm may be used in the program of bread wheat heterosis utilization.
