Cloning and Expression Analysis of Ubiquitin-like Protien Gene in Grimmia pilifera

Abstract: In order to study biological function of ubiquitin-like protein(UBL) in bryophytes,A full-length cDNA sequence of ubiquitin-like protein (UBL) gene was obtained from Grimmia pilifera cDNA library,named Gp-UBL5.The full length of Gp-UBL5 was 471 bp,contained a 222 bp open reading frame(ORF),and encoding a protein with 73 amino acids.Bioinformatics analysis suggested that this protein was a stable protein with a calculated molecular mass of 8.525 kDa and a theoretical pI of 7.84.Phylogenetic analysis indicated that Gp-UBL5 protein had 96% to 99% identities with other UBL5 proteins from different species,such as Brachypodium distachyon and Hordeum vulgare.The qRT-PCR result showed that Gp-UBL5 gene could express in rehydration and dehydration.It was speculated that the gene might play an important role in response to drought.These results provided important information for the further study of functional verification.
