Abstract:
DNA methylation is known to play an important role in the regulation of gene expression in higher?plants. In this study,we assessed the extent and patterns of cytosine methylation alterations in a stable synthetic hexaploid wheat and their progenitor parents using MSAP. Results showed that the levels of methylation in SCA/SQ?(AABBDD) and the parents were 28. 21%,27. 53%,24. 11%,respectively. A total of 1 087 fragments were differentially amplified from the two digests for at least one of the parental lines, about 21. 53% of loci shown changes in?methylation patterns in the allopolyploid. Among these loci,18. 95% were hypermethylated,2. 58% were demethylated. The increased methylation level in synthetic hexaploid wheat compared to its parents might mitigate the shocks?from the hybridization and duplication.
