Abstract: In order to innovate castor germplasm, hulled castor seeds were treated with EMS at 6 levels (0,0.02,0.03,0.04,0.05,0.06 mol/L)and different time(4,8,12 h),The results showed that lethal concentration 50 was at 0.04 mol/L and 4 h,the average taproot length was 10.45 mm,and the concentration of EMS reached to 0.06 mol/L,the seeds could not germinate completely. With the increasing of treatment time and concentration, castor seed germination rate and average relative root length showed a downward trend;ISSR molecular marker analysis showed that DNA variation existed in M2 by EMS.The primer P91 detected 13 variation loci.
