Analysis of Bacillus thuringiensis Strains Containing cry7 Gene

Abstract: Based on PCR amplification using universal primers,18 strains of Bt containing cry7 gene were obtained and analysis further about crystal shape,bioassay,toxin protein and gene type. Bi-pyramidal and irregular crystal shape were presented in these strains. Toxin from Bt strain appeared to contain a prominent protein band at a molecular weight of 130 kDa on SDS-PAGE. Genotypic investigations showed that these strains possessed the cry7 gene, and the amplified fragments were 1 300 bp. Bioassay results showed that the WZ-9 strain was only harmful to larvae of Henosepilachna vigintioctomaculata,c orrected mortality was 93. 1% at 72 h. And the strain WCG-10 showed weak toxicity,corrected mortality was 44. 8% at 72 h. Other strains were no toxicity against this insect. The full length toxin gene,cry7Ab3,was amplified and submitted to the committee of Bt toxin nomenclature (GenBank Number: BI1015188).
