Transformation of Apple Cultivar Gala with a FT Gene Mediated by Agrobacterium tumefaciens

Abstract: In this study,high-frequency regeneration system was founded in leaf blade explants of apple cultiva Gala,which were transformated by Agrobacteria-mediated with FT gene. During cultivating,it was important to keep the anticous surface of leaf not contact with the surface of MS cultivation,after 14d dark cultivation,the regeneration buds and shoots were obtainted in MS (the MS cultivation contained: 5 mg/ L BA,0.5 mg/ L IAA),in this transformation system,the regeneration frequecy was up to 97%. Using cefotaxime sodium (500 mg/ L) and Kanamycin (5 mg/ L) as the in-hibitor of Agrobacteria overgrowth and selection pressure,the explants were infected with Agrobacteria that containted FT gene,then achieved FT gene transgenic plants. The results of PCR amplification suggested that FT gene has integrated into apple plants genomes.
