Study on Bovine Somatic Cell Nuclear Transfer

Abstract: The research was designed to explore a brief procedure on nuclear transfer,which included compare of di-f ferent methods for mature culture of bovine occytes,separation and general culture of fibroblast taken from the skin of bovine,enucleated method and condit ion on fusion which exactly affect the development of reconstructed embryos.Finally the highest mature rate was 81.7% with FSH and LH added,and fusion rate was 93.96% with voltage of 2.6 kV/cm.The cleavage rate and 8-cell development rate of embryos the percentage was 61.2% and 3.27% respect ively when two-half cytoplas-t method was applied,and the data up to 70.8% and 7.56% while blind-cutting manipulation was adopted,and these outcome were all obtained under the condition that reconstructed embryos were cultured in pressing WOW after activation.
