Studies of the Methods for Total RNA Isolation from Plantlets of Apple Cultured in vitro

Abstract: The CTAB-guanidinium method, modified CTAB method, and RNAplant Kit method were used to extract the total RNA from the plantlets of apple cultured in vitro, in which contain a lot of carbohydrates and phenolic compound. The CTAB-guanidinium method showed the best results in term of the quality and quantity of yielded RNA with the A260/A230 value of more than 2.0, A260/A280 ranged from 1.8 to 2.0, and the total RNA yield of 16.95μg/g. The modified CTAB and RNAplant Kit methods were a bit short in total RNA extraction because of a little DNA contamination. And the RNAplant Kit method showed advantages of fastness and convenience.
