Isolation, Identification, Tissue Expression and Bioinformatics Analysis of BMP4 Gene from Binlangjiang Buffalo

Abstract: BMP4 gene plays a vital role in the embryonic development, follicular development and spermatogenesis of human, mouse and cattle, but the research on buffalo has not been reported yet. The purpose of this study was to explore the sequence and tissue expression characteristics of buffalo BMP4 gene in order to reveal its function. The full length of buffalo BMP4 coding region was isolated and identified(KF312880)by the primers designed based on the cattle BMP4 sequence in NCBI database. Then, the molecular characteristics and tissue expression difference of buffalo BMP4 were analyzed using bioinformatics software and semi-RT-PCR technique. The total length of buffalo BMP4 coding region was 1 230 bp, which has the same length with other bovine species, and encoded a peptide of 409 amino acid residues. The buffalo BMP4 protein was a hydrophilic protein without transmembrane domain, containing a signal peptide sequence(AA 1-24)and six kinds of functional active sites. And this protein belonged to extracellular secreted protein, which had a domain of TGF-beta precursor peptide(TGF-beta propeptide)(AA 39-276)and a domain of TGF-beta superfamily(AA 309-409). The amino acid sequence of buffalo BMP4 shared over ≥ 97% homology with that of 10 other species such as wild yak, common cattle, human, horse, etc. Among the 11 buffalo tissues assayed, it was found that the BMP4 expressed only in uterus, ovary and testis, with the highest expression level in testis.The polypeptide encoded by buffalo BMP4 was similar to cattle in structure and function. The results indicated that buffalo BMP4 might play a role in buffalo embryo development and germ cell maturation.
