Detection of Main Causal Viruses on Cucumber by RT-PCR

Abstract: Five pairs of primers were designed and synthesized based on the RNA sequences of cucumber mo-saic viruses (CMV),watermelon mosaic viruses ( WMV一2),tobacco mosaic viruses (TMV),cucumber green mottle mosaic viruses ( CGMMV ), squash mosaic viruses ( SQMV ).The expected fragments were amplified by RT一PCR with the model of total RNA which was extracted from virus-infected and healthy cucumber leaves. A total of 98 cucumber samples associated with viruses collected from several regions of Tianjin in 2002 and 2003 were detected. The results of the experiments showed that the main causal viruses of cucumber in Tianjin were CMV, WMV一2 and TMV with 96. 94 %,77.55%and 38. 67%of incidence rate respectively. CMV and WMV一2 were detected in 76. 53%leaves samples, similarly, CMV, WMV一2 and TMV were detected in 14. 67%leaves samples.
