The Screening of RAPD Markers in V Chromosome of Wheat-hayNa+ldia villosa Substitution Line and Translocation Lines

Abstract: This study screened and analyzed the polymorphism of a wheat-Haynaldia villosa substitution line (6V/6A) and two wheat-Haynaldia villosa translocation lines and parental Haynaldia villosa (VV) and Triticum durum by using 105 pieces of S-series random primers and 100 pieces of Operon random primers. 80.95 percent of S-series random primers amplified the polymorphism and its bands are clear. In all 100 pieces of Operon random primers amplified the polymorphism and its bands can be seen clearly. In 205 pieces of primers, only one primer OPW03 amplified the 570 by band in four materials contained V chromosome of Hay-naldia villosa,but this band don’t exist in common wheat and Triticum durum.Therefore, this molecular marker (OPW03一s}o ) maY locate in the short arm of Haynaldia villosa }s V chromosome.
