Effect of Genotypes and Mediums on Wheat Immature Embryos in vitro Culture

Abstract: The response of immature embryos of 10 wheat genotypes to in vitro cultures on 6kinds of medium was analysed in this paper. The result indicated that doubling the concentration of inorganic solts or adding 1g/1 of NaCl and KCl each on the basis of MS medium cut down the callus induction frequency, while substituting dicamba (4mg/l) for 2,4-D (2mg/l) increased the callus induction frequency. The callus induction frequency of 10 wheat genotypes tested were more than 95%. Callus regeneration frequency varied from 73.7%to 92.5%, and the average was 84.6%. Genotype is a very important factor effecting callus induction and callus regeneration of wheat immature embryos.
