SNPs Detection of IGF-1R Gene and Its Association with Growth Traits in Mink

Abstract: The aim of the paper was to study the single nucleotide polymorphism of Insulin-like growth factor 1 receptor(IGF-1R)gene and its relationship with the growth trait of mink. A total of 235 blood samples from 3 mink varieties(Red-eye white, Jinzhou black and Coffee)were taken as the DNA templates to screen the SNPs by PCR product cloning and Sanger sequencing technology, and the relationships between the mutation sites of IGF-1R gene and the growth traits of different populations were analyzed. The results showed that two SNPs were detected in exon 2 and exon 11 of red-eye white mink and Jinzhou black mink:c.207G > A and c.1782G > A; Two SNPs were detected in exon 2 of coffee mink:c.207G > A and c.218T > A, and one SNP was detected in exon 11:c.1782G > A. Both c.207G > A and c.1782G > A belonged to nonsense mutations, and c.218T>A site belonged to a sense mutation, the later mutation being an amino acid change from proline to arginine. The association analysis showed that the c.207G>A had significant association with the body length of Jinzhou black mink and coffee mink(P<0.05), and GA genotype might be a favorable genotype affecting the body length of mink;while c.1782G>A had significant association with the body length of coffee mink and had extremely significant assosiation with the body weight of coffee mink(P<0.01). The heterozygous genotype(GA)was the dominant genotype, and its body length and body weight were significantly higher than GG genotype and AA genotype individuals. The association analysis of the combined genotypes showed that the different genotypes of Jinzhou black mink were significantly correlated with the body length and body weight. The body length of the combined genotype GGGA individuals and GGAA individuals were extremely significantly higher than other genotypes (P<0.01); The body weight of the combined genotype GGGA individuals were significantly higher than GAGA genotypes individuals (P<0.05); It was preliminarily concluded that the GGGA and GGAA genotypes might be a favorable genotype affecting the body weight of Jinzhou black mink. The above results indicated that IGF-1R gene might be the candidate gene affecting the growth trait of mink.
