Polymorphism of SNP Locus at GH Gene in Finland Blue Fox and Local Blue Fox

Abstract: The research was to study the relationship of the polymorphism of GH gene on body measurement traits in blue fox gene,providing some evidence for the selection and breeding in blue fox gene. Finland blue fox and local blue fox with significant differences in body weight and body length were chosen for testing subjects,DNA pooling was constructed for each species. The technology of direct sequencing of PCR products was used to detected the single nucleotide polymorphism of fox growth hormone gene. And correlation analysis between GH polymorphisms and growth traits was carried out. Results showed that two SNPs,A306G and C1144T were found within finland blue fox and local blue fox.The genetic structure analysis demonstrated that the two SNPs loci in the finland blue fox were low polymorphism loci (0.057 9) and local blue fox were high polymorphism loci (0.573 1). The two SNPs loci were the same in the genetic diversity parameters,the frequencies of alleles and genotypes,thus had complete genetic linkage,with the formation of AACC,ABCD and BBDD. The analysis of variance showed that the gene frequency difference among finland blue fox and local blue reached at the extremely significant level(P <0.01). Moreover,we analyzed the correlation between the genotypes of SNPs loci and the growth traits of the two groups.The male fox body weight of AACC genotype were significantly higher than those of ABCD genotype(P <0.05),but there was no significant difference in body length (P >0.05).The female fox body weight of AACC genotype were extremely significantly higher (P <0.01) and body length significantly higher than those of ABCD genotype(P <0.05). These results indicated that the two SNPs might be related to the growth performance.
