Fingerprint Construction and Genetic Diversity Analysis of Sorghum ×Sudan Grass by SRAP Markers

Abstract: The DNA fingerprint of 22 Sorghum Sudan grass varieties was established by using Sequence-Related Amplified Polymorphism markers(SRAP),with 99. 9999% probability of confidence. Using 19 SRAP primer combinations which were selected with abundant polymorphic to analyze genetic diversity among 22 Sorghum Sudan grass. A total of 450 bands were scored by the primer,373 out of them were polymorphic,with an average of 23. 6 bands and 19. 6 polymorphic bands for each primer combination. The percentage of polymorphic loci was 82. 9%. According to the genetic similarity coefficient among different strains and Un-weighted Pair-Group Method Arithmetic Average (UPGMA),Genetic similarities among the 22 Sorghum Sudan grass varieties ranged from 0. 43 to 0. 93 with an average of 0. 66. With the similarity coefficient criteria of 0. 674 these 22 accessions were clustered into 2 major groups. The cluster analysis showed that cluster I could be classified into 3 sub-clusters. The results verified very high discrimination ability of SRAP markers,moreover,considering its simple,stable,codominant and well-proportioned distribution,conclusion could be made that SRAP might be a potentially useful marker technique and supplement for SSR in Sorghum Sudan grass hybrid identification. The fingerprinting pattern produced by this study could be applied for identification of the studied genotypes.
