Analysis of Genetic Diversity Core Collection of Tree Peony Cultivars from Central China Using SRAP Markers

Abstract: In order to explore the genetic diversity of core collection of tree peony cultivars from central China, the technology of SRAP molecular marker was used in the study.Based on the technology of SRAP molecular marker, the genetic diversity of the core collection of tree peony cultivars from central China.The results showed that 23 SRAP primer combinations were employed to evaluate the genetic diversity of the core collection including 58 tree peony cultivars, from which a total of 595 amplified fragments were detected, and 377 of them were polymorphic with a polymorphism percentage of 64.3%, the number of amplified fragments and the number of polymorphic bands for each primer combinations was 25.9 and 16.4, respectively.Its genetic similarity coefficient in pairs was between 0.567 and 0.894, the average was 0.752.The 58 tree peony cultivars were grouped into seven clusters at genetic similarity of 0.692.This indicated that the genetic diversity of these cultivars was abundant.Moreover, the clustering results showed that the cluster was not completely consistent with the classification of floral forms though there was a tendency that the single-flower section and the proliferate-flower one were to get together separately.And the cultivars with close color scattered in different categories instead of clustering together.That is to say there is a great genetic difference between the low and high polyphyll cultivars, but there is no clear relationship between the clusters with floral form, color and characteristics.This might be due to the introduction, selection and repeat hybridization of tree peony for a long time, leading to the complicated sources of varieties
