Identification of Seed Prolamin in Foxtail Millet from Difference Zones(Setaria italica L.Beauv.)with SDS-PAGE

Abstract: Prolamin is the main storage protein of foxmail millet, and it is meaningful for the research of prolamin polymorphism in millet breeding and genetic resource evaluation.With SDS-PAGE, prolamin polymorphism of 46 foxtail varieties from different province was analyzed.The results indicated that 14—24 prolamin bandings could be identified from one variety。and each variety contained 18.74 bands on average.There were 32 bands in 46 varieties, 25 of them showed polymorphism(amount to 78.1%)and 7 of them were shared by all the varieties.Prolamin bands could be divided into A, B, C and D sections based on mobile rate, and there were 6, 16, 15, 4 band patterns in four sections, respectively.Ppolymorphism is rich in B and C section.Bands in A and B sections were relatively weaker, and that stronger in C and D sections.The molecular weight of prolamin was 13.0—55.0 kDa.The prolamin bands around the 29, 25 kDa(No.23), 22.5 kDa(No.26), 17.5 kDa(No.28)sites, were relatively stronger, and significantly different among varieties.The research result could be used in variety identification and evaluation.The cluster analysis of prolamin indicated that 46 varieties fallen into five categories based on the GD value at 0.45. in which 34 varieties were in the same categories and the other four categories only contained a few varieties.The resuits indicated that most of the materials were of high similarity.
