Genetic Diversity Analyzed by EST-SSR Marker and Polymorphic Traits of 31 Paeonia lactiflora Cultivars

Abstract: Genetic diversity of 31 Paeonia lactiflora cultivars was analyzed by EST-SSR marker and phenotypic traits.Eighty EST-SSR primers from Peony were screened against genomic DNA of Xueshanjinhui,Zhencilian,Biandihong and Zitanshengyan,and 52 primer pairs showed the polymorphism,accounting for 65% of total primers.Then the 52 EST-SSR primers showing polymorphism were subjected to reveal the genetic relationships among the 31 Paeonia lactiflora cultivars.The results showed that among the 31 Paeonia lactiflora cultivars,52 EST-SSR primers pairs had 265.2 polymorphic loci,and 5.1 polymorphic loci were detected by each primers pairs.The Nei's distance between cultivars was calculated for cluster analysis.Moreover,the genetic distance based on the agronomic characters,such as flower color,flower form,plant height,plant width,flower diameter,leaf length,blade width,plant type and leaf type was calculated by Euclidean distance.The classification of Paeonia lactiflora cultivars using the Unweighted Pair-group Method Arithmetic Averages (UPGMA) based on the pair distance is mostly consistent with known pedigree information.The 31 Paeonia lactiflora cultivars have been mainly classified into six groups. GroupⅠconsists of Qihualushuang, Zhushapan, Cibai, Yanzhidianhong, Yulinhong, Yinlonghuanzhu, Moziling, Ziyunu,Baiyubing and Ziliantuojin; Group Ⅱ consists of Hongyanfeishuang, Tianshanhongxing, Zitanshengyan and Xueshanziyu; Group Ⅲ consists of Huahonglou and Zifengyu; Group Ⅳ consists of Zhencilian, Pantuoronghua, Baiyupan and Fenchidicui; Group Ⅴ consists of Biandihong, Tuopanxianbao, Dongfanghong, Ziyanfeishuang, Meiguipiaoxiang and Xueshanjinhui; Group Ⅵ consists of Zihongkui, Hongjuhua, Hongpantuojin and Duoyezi. Mantel test revealed that the goodness of fit between EST-SSR marker and phenotypic analyses was significant (r=0.802 3,t=10.324) in analyzing detecting genetic diversity of the cultivars.This study provided theoretic base for new cultivar breeding and industrialization of Paeonia lactiflora in the future.
