Detection of a RAPD Marker Linked to rht Gene in Wheat (Triticum aestivum L.)

Abstract: Four near-isogenic lines which contained rht,Rht1,Rht2,Rht3 respectively were used for polymorphic analysis at molecular level by RAPD technique. Out of 296 10-mer oligonucleotide primers screened, 25 showed polymorphism among near-isogenic lines. During 6 repeat tests of these primers, 18 could not give the repeat results, 6 could repeat 2 to 3 times, only primer OPAM01 showed the stable results in which a 1.86 kb additional band was found in rht line. OPAM011860 could be identified as a RAPD marker linked to rht gene in wheat.
