Studies on the Expression of Foreign Thaumatin-like Protein Gene in Potatoes

Abstract: Tuber discs of "Jinyin potato No 8" were co-cultivated with an agrobacterium tumefaciens binary vector, carrying both the thaumatin-like proteins gene under the control of the CaMV 35S promoter and the bar gene as a selectable marker under control of the ubiquitin promoter. Tuber discs started to form a number of shoots on shoot inducing medium two to three weeks after co-cultivation. The shoots were inoculated on MS medium containing the herbicide bialaphos. The potato seedlings which grew well on MS medium containing bialaphos were assayed by indirect ELISA. The positive samples were assayed by PCR to see the existence of the TLP gene. The results of it indicated that the foreign thaumatin-like protein gene was introduced into the potato genome and expressed in the transgenic potato.
