Production of Antibody against Wheat Native Calmodulin and Its Application to Enzyme Labelled Immunohisto-Chemistry

Abstract: Calmoduin (CaM) from wheat coleoptile was purified and unmodified. Native CaM was used to induce antibody against wheat CaM. Assay of immunological characteristics of the antibody obtained showed that the antibody against wheat CaM almost could not cross-react with bovine brain CaM, but could do with CaM from a number of kinds of plants. This showed that antibody obtained was specific for CaM from plants. Localization in cultured tabacco cells with enzymelabelled immunohistochemistry showed that there was more CaM in cytoplasma, but the nuclei, especially nucleoli, also contained CaM. The significance of localization was discussed in the paper.
