Effect on the Na+ Tolerance of Yeast Transformed with Wheat HKT Genes Family

Abstract: The members of HKT family play an important role in maintaining Na+/K+ homeostasis.In order to analyze the role of wheat HKT genes in salt-tolerance mechanism,comparative analysis on Na+ affinity of wheat different members,the copies on different genomes and the member's alleles of wheat HKT family were conducted by transforming yeast mutant G19.Three members of HKT family, HKT1; 5, HKT2; 1,and HKT2; 2,showed different Na+affinity.Na+affinity of HKT2;2-A was higher than HKT2;2-B.Leu48→Pro48 mutation increased Na+ affinity while His215→Tyr215 mutation decreased that of HKT2;1.The results indicated that the two amino acids may be related to Na+ transporter in common wheat.
