Genetic Diversity of Leuciscus waleckii Populations Based on CO ⅠSequence

Abstract: To analyzing the genetic diversity and genetic structure of Leuciscus waleckii based on mtDNA CO Ⅰgene(635 bp length),six geographical populations(72 samples) were sampled from Dali Lake(DL),Ganggeng Lake(GG),Hulun Lake(HL),Amur River(HLJ),Songhua River(SHJ) and Wusuli River(WSL).On the base composition,A+T content(52.37%) was slightly higher than that of G+C content(47.63%),corresponding to the characteristics of nucleotide composition in the vertebrates;On the genetic parameters,the genetic diversities of Dali Basin populations(DL and GG) were lower than those of Amur Basin counterparts(HL,HLJ,SHJ and WSL);There were 20 haplotypes,of one haplotype was shared by all the six populations,one shared by Dali Basin and one shared by Amur Basin.Despite there were significant population differentiation between Dali Basin and Amur Basin(P<0.05) as well as genetic structure,the analysis of AMOVA and the values of frequent gene exchange(Nm) both demonstrated that the six Leuciscus waleckii populations had very close relationship,probably derived from a common ancestor,and had not yet differentiated to different species or subspecies based on present evidence.
