Cloning of Wheat TaWRKY46-1 Gene and Construction of Plant Inducible Expression Vector by Gateway Cloning Technique

Abstract: In this study, the wheat variety, Y anon ai 158(Triticun aestiven L),was used as the experinentalm,terial for cloning the wheat TaWRK Y46 gene Primers were designed based on a known wheat TaWRKY46 gene sequence in NCBI, subsequently an 870 bp cDNA fragment was amplified from total RNA of Yarm ai 158 by RT-PCR Sequence analysis revealed that the cloned fragment contains the full-length cDNA and is highly hologous to the TaWRKY46 sequence in NCBI The sequence alignment revealed a Q to E change with in the conserved "WRKYGQK" domain, suggestilg it was a new allele of TaWRKY46. Hencewe designated it asTaWRKY46-1. In onler to e1uciclate liznctional role of the new Yangnai 158TaLYRKY46-1 allele, a plant ioucible over expression vector, pKIGW-TaWRKY 46-1, was constmcted via Gatrway cloning technology and tr-anslonned intoAgroFxzcteriun cells successiiilly Thiswmk proviles the key basis for future detailed functional analysis of wheat TaWRKY46-1 gene.
