Identification of Apple Rootstocks with AFLP Molecular Markers

Abstract: Thirty apple rootstocks were subjected to AFLP analysis.Three primer combinations with high polymor 2 phism and powerful distinctiveness selected from 64 primer combinations produced 199 fragments,of which 176 were polymorphic with a polymorphism percentage of 88.4 %.The results showed that characteristic bands were amplified from 12 rootstocks,and any of the three primer combinations could identify all the rootstocks.AFLP was proved to be a very reliable method to identify apple rootstocks.The genetic relationship of the apple rootstocks was analyzed by clustered analysis,which provided the scientific basis for further collection and the use of the Chinese fine rootstock resources.
