Isolation and Characterization of a Lea3 Gene Fragment from Agropyron mongolicum Keng

Abstract: In this study,drought resistance gene homology fragment fromAgropyron mongolicumKeng was isolated using homology-based method.A pair of degeneracy primers was designed according to the amino acid conserved regions of the clonedGramineaeplant group 3Leagenes,One drought resistance gene has been obtained.by the polymerase chain reaction(PCR)using the genomic DNA of A.mongolicumKeng as template.It was named asLea3.The nucleotide sequences was 497 bp,including 124 bp untranslated region,and encoded 124 amino acids.Homology research showed that the nucleotides ofLea3 was 93 % identical to wheat group 3Leagene Wrab19,93 % identical to wheat gene WRAB1,96 % identical to the clone 9124 from the mRNA of maize,94 % identical to barley genepHVA1,and 91 % identical to barley geneHVA1.the analysis of its secondary structure and hydrophilicity indicated that 2-helixes were predominant feature of secondary structure,and this gene had high hydrophilic property.The result of Southern blot analysis revealed the existence of anLea3 gene family in the A.mongolicum Keng genome.
