Polymorphism Analysis inBMP15Gene Extron1 Site and Its Association with Prolificacy in Mongolian Sheep

Abstract: The single nucleotide polymorphism(SNP)at extron1 site ofBMP15gene of Mongolian sheep was studied by PCR-SSCP and its association with prolificacy was analyzed.The results indicated that theBMP15geneshowed poly-morphism in the 116 individuals and the genotypes++,A+and AA were found in Mongolian sheep.Sequencing analysis indicated that the 28th,29th,30th base in the encoding domain were lost in the DNA sequence,and the 10th amino acid residue of leucine(L)was lost by the mutation in the Wild Type(++)amino acid sequence,forming the B1 mutant and the genotype was AA.Population genetic analysis indicated that the B1 mutat ion occurring in twins lamb strain was higher than in single lamb strain,x2test indicated that the two strains were in moderate polymorphism and at Hardy-Weinberg equilibrium(p<0.05)in this site.The B1 mutation had no significant effect on the litter sixe by analyzed the association of the polymorphism with high prolificacy in Mongolian Sheep.
