Identification of Molecular Markers for Wheat Stripe Rust Resistance Gene Yr6

Abstract: Wheat stripe rust resistance gene Yr6 has been used very limited in wheat breeding in China, identification of molecular markers for Yr6 gene will promote reasonable use of the gene. Combining Yr6 gene with other effective stripe rust resistance genes will enhance stripe rust resistance and prolong commercial life of the cultivars bred. In this study, DNA polymorphism between the near iso-genic lines Yr6/6×Avocet S and susceptible line Avocet S was analyzed with 653 RAPD primers and 36 pair of SSR primers. The SSR primers with polymorphic PCR fragments were tested further on 2 Yr6-carring cultivars Heines Kolben and Heines Peko. The results showed that 93 (14.2% of total) RAPD primers and 5 (13.9% of total) pair of SSR primers amplified polymorphic DNA fragments between Yr6/6×Avocet S and Avocet S. The primers of SSR markers Xwmc76 and Xwmc276 amplified the same bands in the 2 Yr6-carring cultivars as in Yr6/6×Avocet S but different in susceptible line Avocet S, which implied the 2 SSR markers maybe linked to Yr6 gene.
