Molecular Mapping of a Powdery Mildew Resistance Gene Transferred from Wild Emmer into Common Wheat

Abstract: Wheat line 7K65 was resistant to Blumeria graminis f. sp. tritici Race E09. Genetic analysis showed that the resistance was controlled by a single dominant gene,named MlTK65. After screening SSR,EST-STS markers,we found 7 marker loci were linked to the powdery mildew resistance gene Ml7K65. The genetic map of gene Ml7k65 indicates the order of these marker loci is Xcfa2086-XEST83-XEST89-Xksum193-Xmag633-Xcfd50-Ml7K65. The genetic distances of each interval are 5.4,2.8,0.4,2.4,0.4,2.0 cM, respectively. Xcfd50 is the closest marker, the genetic distance is 2.0 cM,while Xcfa2086 is the most distant maker with the genetic distance of 13.4 cM. Using Chinese Spring nullisomic-tetrasomic lines, we physically located the markers linked to Ml7K65gene on wheat chromosome 2A.
