Optimization of Agrobacterium tumefaciens Mediated Genetic Transformation System of Mopanshi

Abstract: ACC synthase gene was transferred into Mopanshi mediated by Agrobacterium tumefaciens using young leaf of Mopanshi as explant. Effectors that influenced transformation were studied and the optimum genetic transformation system was established. The results showed that 2 d of pre-cultured time, 1.0 OD600 of engineering bacterial suspension, 15 min of infection time, 3 d of co-cultured time and 200 μmol/L of AS were optimum for increasing the frequency of transformation. It was proved that the ACS gene had been transferred into Mopanshi by PCR.
