RFLP Analysis on Mitochondrial Cytochrome Oxidase I Gene of Chinese Mitten Crab (E. sinensis)

Abstract: The fragments of mitochondrial cytochrome oxidase I of Eriocheir sinensis from Changjiang, Oujiang and Liaohe were amplified by using PCR-RFLP technique. 11 restriction endonucleases were used to digest the PCR products. In this study there were 6 composite haplotypes in the 3 populations, there were 5 composite haplotypes in Oujiang population and 2 ones in population of Liaohe and Changjiang. The index of nucleotide diversity (π value) were 0.0167 (Oujiang), 0.0067 (Changjiang) and 0.0003 (Liaohe) respectively, indicating that the genetic diversity of Oujiang population was highest among the 3 populations, the genetic diversity of Changjiang population was higher than that of Liaohe population. Interpopulational net genetic distance was the highest (0.0237) between Liaohe and Oujiang, while it was 0.0207 between Changjiang and Oujiang and was 0.0043 between Chanjiang and Liaohe samples. The results indicated that the relationship between the populations from Changjiang and Oujiang was the closest, and that of Liaohe and Oujiang populations was the most distant.
