Effect of Water Stress on the Winter Wheat Callus

Abstract: With immature embryo cultured in vitro on the modified medium MS, the callus of two common chinese winter wheat, drought-resistant, Hua 84, and drought-susceptible, JiMai 24, were induced in dark. After subcultured on the same component subculture-medium two times (each subculture period was 30 d or so), they were cultured in two kinds of liquid mediums, which contained different concentration of PEG-6000 (10%, 20% W/V) so as to induce different intensity of water stress for 24 hours. The callus of the two winter wheats were studied and were compared with each other. The results showed that. It was found that the level of free proline, the activity of POD, the plasmolmma permeability increased under water stress, while the tissure vigor decreased. In all experiments, the drought resistance wheat, Hua 84, was more superior to the drought-susception wheat, Jimai 24. It showed more vigorous under water stress. Drought resistance at the cellular level was accord with that at the intact plant level. So the callus can be used to study the mechanism of drought resistance instead of intact plant.
