Isolation and Identification of H1N1 Subtype of Swine Influenza Virus

Abstract: Three strains of swine influenza virus(SIV)were isolated from pig farms in Tianjin region in 2005 to 2006.All of the three isolates showed hemagllutinin activity to l%RBC of cock and were positive in AGP test with antibody of SIV Systemic identification was done by China Animal Healthy and Epidemiology Center and the result showed the three isolates were belong to H1Nl subtype.The isolates were christened as A/Swine/Tianjin/TJ2/2005 (H1N1),A/Swine/Tianjin/TJ3/2005(H1N1)and A/ Swine/ Tianjin/TJ4/20(O(HI N1).All of the isolates of SIV were became uninfected to chicken embryo in 60℃water bath 10 rain.pH3.0 incubated 3 h at 25.C and incubated overnight in 4.C with aether.EID卯of the three isolates were 10-7-//02mL and 10-8-/0.2 mL respec. tively.The isolates of SIV couldn 7t cause disease in chicken and mice after infected,but some of pigs were get sick post inoculated in all group infected by the three isolates and Hl N1 subtype of SIV were gotten from lungs of infected pigs.
