Isolation,Identification and Biological Characterization of Swine Influenza Viruses in Henan

Abstract: Samples were collected by cotton swabs and lung tissues from pigs with respiratory system problems on 5 farms in Henan province. Samples were inoculated into 10-day-old SPF-embryonated eggs via the allantoic route,after being processed. Three isolates were obtained and identified as influenza A virus subtype H3N2 by HA and HI. Some biological properties of them were studied. 50% egg infective doses (EID50) were 10 -9. 56 /0. 2 mL,10 -8. 56 /0. 2 mL and 10 -8. 44 /0. 2 mL. 50% lethal doses (LD50) to small white mice of the strain were 10 -1. 56 /0. 1 mL,10-1. 45 /0. 1 mL and 10 -1. 50 /0. 1 mL ,respectively. Hemagglutinins of the strains were unstable to heat. Red blood cells of horse, cattle,pig,donkey,pig,sheep,cock,rabbit,guinea pig,small white mouse,big white mouse and human O-type could be agglutinated by the isolate. It will lay a foundation for understanding influenza viruses circulating in swine herds in Henan province.
