Effects of Porcine Circovirus-like Agent P1 Infection on the Porcine Antiviral Protein mRNA Transcriptions in Peripheral Blood Mononuclear Cells of Piglets

Abstract: The study was to explore the effects of porcine circovirus-like agent P1 infection on the porcine antiviral protein mRNA transcriptions. About thirty-day-old healthy Guangxi Bama miniature pigs were inoculated with cloned genomic DNA of P1. The changes of mRNAs of porcine antiviral protein (PKR,2'-5' OAS2,RNase L and Mx1) in peripheral blood mononuclear cells(PBMC) of piglets were analyzed by real-time fluorescent quantitative PCR (real-time FQ-PCR). The results showed that the transcriptions of PKR mRNAs were increased in the affected piglets at 8 and 40 day post-inoculation(DPI), respectively. Whereas 2'-5' OAS2 mRNA levels of the inoculated pigs were downregulated at 3 DPI and 17DPI,the P1 infected group trended to decrease expression of Mx1 mRNA at 17DPI as well. RNase L mRNA expressions appeared to be no significant change by P1 infection. In conclusion,the results indicated that P1 infection can affect the mRNA levels of PKR,2'-5' OAS2 and Mx1.
