Identifying the Homoeologous Groups of the Alien Chromosomes of Triticum aestivum-Leymus multicaulis Disomic Chromosome Addition Line Line24 Using SSR Markers

Abstract: Of the 227 wheat microsatellite prime pairs used to amplify products from Leymus multicaulis, 76 showed polymorphisms among Leymus multicaulis, the common wheat parents Chinese Spring and Fengkang No. 13 of the T riticum aestivum-Leymus multicaulis disomic chromosome addition line Line 24 which was tolerant to Barley Yellow Dwarf Virus ( BYDV). These 76 primer pairs can be useful for identifying the homoeologous groups of the alien Leymus multicaulis chromosomes of Line 24. Among them, 4 could am- plify same product from Line 24 and Leymus multicaulis, but different from Chinese spring and Fengkang No. 13. The same result was obtained when using 7 different monosomic chromosome addition lines whose parents were Line 24 and different common wheat varieties as template. Therefore, these 4 microsatellites could be used as molecular markers for the alien Leymus multicaulis chromosomes of Line24. According to the loci of the products of those 4 microsatellite prime pairs in wheat, it can be concluded that the Leymus multicaulis chro- mosomes of Line 24 might contain the fragments of 3Xm( Ns), 5Xm( Ns), 6Xm( Ns) and 7Xm( Ns) chromo- somes of Leymus multicaulis. They must have exchanged chromosome fragments each other.
