Transcriptomics and Proteomics Correlation Analysis of Fatty Acid Metabolism in High Oil Rape

Abstract: In order to study the regulation mechanism of fatty acid metabolism in high oleic rapeseed,a group of highly oleic acid near-isogenic lines was used as the material from 20 to 35 days after pollination,and was discussed by isobaric tags for relative and absolute quantitation with transcriptome technology. The results showed that the correlation coefficient between protein and transcript group was not high,and the correlation coefficient between quantitative protein and gene was-0.326 9. The correlation coefficient of differential protein and gene expression was-0.735 8,and the correlation coefficient of identical protein and gene expression was 0.714 3. Besides,80 differentially expressed genes were identified,GO annotation suggested that these genes involved in metabolism,signal transduction,defense and stress responses,redox,transcription,and so on,23 of the 80 genes were associated with fatty acid metabolism,16 of them were up-regulated and 7 down-regulated. gi|297330358(pantolactone ACP methyl ester),gi|297321940 (phosphorylase)and gi|297314818 (acetamide/formamidase family) may play an important role in the case of high oleic acid rapeseed fatty acid metabolism.This study shows that proteomics and transcriptase association analysis can provide a new idea for the mechanism of fatty acid metabolism in high oil rape.
