Genetic Analysis and Molecular Mapping of a Stripe Rust Resistance Gene YrCH5026

Abstract: CH5026 is a stable introgression line carrying resistance genes of Thinopyrum intermedium.To better utilize CH5026,increase the genetic resources,this study were to characterize the origin of the resistance,determine the inheritance of resistance,locate the chromosome and construct the linkage map.Using three prevalent races of stripe rust CYR31,CYR32 and CYR33, Th.intermedium (accession Z1141),CH5026 and its resistant parent amphiploid TAI7045 as well as all wheat parents were evaluated by inoculated races in the seedling stage and adult stages.The result showed that CH5026 and its donor TAI7045 as well as the wild parent were resistant at the seedling and adult stages to CYR31,CYR32 and CYR33,whereas all the wheat parents involved were susceptible,indicating that the CH5026 was possibly derived from Th.intermedium. No Th.intermedium chromatin has been detected according to genome in situ hybridization assay.Genetic analysis on the F1,F2,F2:3 and BC1 populations from stripe rust resistant line CH5026 revealed that a single dominant gene resistance to CYR32.A total of 192 F2 populations from the cross CH5026/Taichang 29(TC 29)was screened with 569 wheat SSR primer pairs to detect molecular markers linked to resistance gene.Three SSR primers, Xgwm210, Xwmc382 and Xgpw7101 were identified to be linked to the resistance gene.The two flanking markers were Xwmc382 and Xgpw7101,genetic distances were 6.0,4.7 cM,respectively.Using the Chinese Spring(CS)nulli-tetrasomic and ditelosomic lines,the resistance gene and its linked markers were assigned to chromosome arm 2AS.Based on the source of the Yr genes and ploymorphism of linked molecular markers,the stripe rust resistance gene in CH5026 was likely different from the known resistance gene on chromosome 2AS.The gene might be a novel stripe rust resistance gene,this new resistance gene is temporarily named YrCH5026.
