Multiple PCR Detection of Fusarium oxysporum and Fusarium solani in Cucurbitaceous Plants

Abstract: The early symptoms caused by F.oxysporum and F.solani were similar in cucurbitaceous plants such as watermelon.To rapidly and accurately distinguish the two diseases at a potential or early infection,three primes were designed based on the TEF1-α(Translation elongation factor 1-α) of F.oxysporum and F.solani. The primer pair FO-F+Fu-R amplified a 228 bp fragment from F.oxysporum,and the primer pair FS-F+Fu-R amplified a 347 bp fragment from F.solani.No PCR products were amplified with these primer pairs from DNA of other fungal species.A multiple species-specific PCR method was developed successfully to detect both two fusarium fungi simultaneously in single PCR amplifications with the primers FO-F+FS-F+Fu-R.
