Cloning of Small Molecular Shock Protein Gene Nt-Hsp17.8 from Nicotiana tabacum and Its Expression Analysis under Inuced by Stress

Abstract: Increasing evidences show that abiotic stresses can affect plant growth and development and the range of cultivation.In order to increase cultivating resistance of tomato and potato in the combination of heat and drought stresses through transgenic methods,cytoplasmic class I sHSPs cDNA with full length of 876 bp was cloned from heat-shocked leaves of Crofton weed(Nicotiana tabacum L.) by conducting RT-PCR and RACE-PCR.Sequence analysis showed that it included a 480 bp open reading frame(ORF) which encoded 159 amino acids with a deduced molecular weight of 17.8 kDa.It was called as Nt-HSP17.8.The results of Northern blot showed that expression of Nt-HSP17.8 gene in tobacco leaves was induced not only by a single stress such as heat or drought,but also by combination stress(heat shocK+ drought) conditions,indicating that the gene might play an important role in the combination of heat shock and drought stress.
