Study on Screening and Identification of Cellulolytic Strain LY16 and Its Cellulase-producing Conditions

Abstract: Samples were collected from rotten wood and soil nearby, and 25 strains that can decompose the cellulose were separated, among which the strain LY16 has the strongest cellulase activity. It was identified as Trichoderma reesei by morphological observation and the molecular biological methods. By carrying out the carbon-nitrogen source and other experiments, we have obtained the optimal conditions in which this strain produces enzyme. The culture compositioon (g/L) contained :avicel 10, wheat bran 40, peptone 4, urea 12, KH2PO42, MgSO4 ·7H2O.1, CaCl2·2H2O.1, FeSO40.01, MnSO40.004 5, CoCl20.0036, ZnSO40.0035, Culture temperature was 30 ℃, Liquid volume in flask was 50 mL, rotational speed was 200 r/min, culture time 72 h. In this condition, the CMCase was 202.6 U/mL, and the FPase was 53.2 U/mL.
