Application of AgNO3 to Someclone Culture of Immature Embryo in Wheat

Abstract: In order to improve the regeneration frequency of somatic embryos of wheat immature embryo,the effects of different concentration AgNO3 on someclone culture of immature embryo callus and oxalate oxidase activities of immature embryogenic callus were studied in this paper from three wheat cultivars (Yumai 18,Yumai 49 and Lankao 906) which were cultured with more areas in Henan province.The result indicated that the differences of induction frequency of emryogenic callus andgreen spot rates of callus and oxalate oxidase activities of immature embryogenic callus were significant at 0.01 level among different AgNO3concentrations or genotypes.It was highest in Yumai 18 and was lowest in Lankao 906 among different genotypes.It were highest in MS medium suppliedwith 2.55.0 mg/ L AgNO3 among different AgNO3 concentrations.The coefficient between induction frequency and oxalate oxidase activities of immature emryogenic callus in wheat under different AgNO3concentrations was0.920 and it was significant at 0.01 level.So,the regeneration frequency can be increased in someclone culture of wheat immature embryo supplied with Ag+.
