Study on Isolated-microspore Culture and Plant Regeneration of Cauliflower

Abstract: The cultivation of isolated microspores with 186 caulfflowers cross cultivate and combinations were studieb. The results showed that genotype was a critical factor affecting embryogenesis in isolated microspore culture. The heat shock pretreatmern at 32 ℃ for 24 h was necessary for embryo formation; the stage from uninuclear to binuclear was the optimum period for embryo inocanent and the highest frequency of anbryogenesis could be attained; it also indicated that the addition of 0.1 mg/ L CUBA and 0.005 mg/L NAA to culture medium could promote embryoid development. The survival rate of 20-day embryo was greatly enhanced when treatebwith dedifferentiation. Consequernly, the ratio of plant re-generation was 53%, the ratio of DHin regenerate plarn was 83.5%.
